Prunus persica has been proposed as a genomic model for deciduous trees and the Rosaceae family. Optimized protocols for RNA isolation are necessary to further advance studies in this model species such that functional genomics analyses may be performed. Here we present an optimized protocol to rapidly and efficiently purify high quality total RNA from peach fruits (Prunus persica). Isolating high-quality RNA from fruit tissue is often difficult due to large quantities of polysaccharides and polyphenolic compounds that accumulate in this tissue and co-purify with the RNA. The aim of this thesis is to improve knowledge on mechanisms involved in the response to nitrogen limitation and in lipid accumulation in the microalgae haptophyte Tisochrysis lutea. The wild type strain and a lipid accumulating mutant strain were grown under different nitrogen limitation and starvation and analyzed by functional genomics. Four genes of high-affinity nitrate/nitrite transporter (Nrt2) were identified and characterized to reveal the mechanisms involved in mineral absorption in this species. Transcriptomes of both strains were sequenced and proteins affected by nitrogen starvation and differentially expressed between the two strains were identified. The oyster Crassostrea gigas has global distribution and for the past several years the highest annual production of any freshwater or marine organism. Abstract Genes related to the response to drought stress in leaf and root tissue of drought-susceptible (DS) and tolerant (DT) genotypes were characterized by RNA-Seq. In total, 54,750 transcripts, representative of 28,590 genes, were identified; of these, 1,648 were of high-fidelity (merge of 12 libraries) and described for the first time in the Andean germplasm. From the 1,239 differentially expressed genes (DEGs), 458 were identified in DT, with a predominance of genes in categories of oxidative stress, response to stimulus and kinase activity. Most genes related to oxidation-reduction terms in roots were early triggered in DT (T75) compared to DS (T150) suggestive of a mechanism of tolerance by reducing the damage from ROS. The field of Genetics is undergoing tumultuous change after nearly a century of standard approaches to genetic analysis. The Human Genome Project is providing tools and technologies that are changing the ways that we pursue an understanding of gene function, which is the underlying goal in modern and traditional genetics. In this paper, I overview the directions of the genome project as they relate to gene function analysis in mice and humans, and how various modern technologies are coalescing to address this in a powerful way.
